Single-cell atlas maps immune and metabolic shifts in testicular mixed tumors

Researchers have built a single-cell transcriptomic atlas of testicular mixed germ cell tumors (TMGCTs), a rare but increasingly prevalent subtype of testicular germ cell tumors whose cellular heterogeneity and microenvironment have remained poorly understood. The study, published in Cellular Oncology as an open access article, identifies 13 major cell types within these tumors and documents marked immune infiltration alongside a loss of spermatogenic lineages.

A central finding concerns the malignant cells themselves. The team subdivided them into three distinct subclusters, each carrying its own unique transcriptional program. Pseudotime trajectory analysis suggested a developmental hierarchy among these malignant populations, a structure supported by the dynamic expression of pluripotency factors such as POU5F1.

The analysis also pointed to extensive metabolic reprogramming in the tumor cells. In particular, the researchers uncovered a significant upregulation of the N6-methyladenosine (m6A) RNA modification machinery within the malignant compartment. Two components of this landscape, METTL16 and POU5F1, showed prognostic relevance, marking them as candidate biomarkers worthy of further investigation.

Within the tumor microenvironment, macrophages emerged as a dominant presence. The atlas revealed a predominance of MRC1+ macrophages exhibiting pro-angiogenic features, suggesting a role for these immune cells in supporting the tumor’s vascular supply.

The T cell compartment proved equally distinctive. The researchers identified distinct T cell subsets, including a notable presence of effector T cells, naïve T cells and regulatory T cells.

Vascular cells within the tumors showed their own specialized program. Tumor endothelial cells were enriched in tip cell and vein populations and exhibited high expression of angiogenesis-related genes as well as genes involved in collagen formation, underscoring an actively remodeling vasculature and extracellular matrix within the tumor.

One extracellular matrix gene stood out prognostically.

Taken together, the authors present the work as a comprehensive single-cell transcriptomic atlas that reveals the cellular ecosystems and transcriptomic drivers of TMGCTs, and highlights potential biomarkers and therapeutic targets for this understudied tumor type. Because the findings derive from a single-cell mapping study, they identify associations and candidate targets rather than validated treatments, and clinical application would require further validation.

The study is currently available as a peer-reviewed accepted manuscript shared early for faster access; this citable version carries a permanent DOI and remains subject to further edits before the final Version of Record replaces it.
