Review Explores Metabolic Reprogramming of Regulatory T Cells in Colorectal Cancer

In a review published in the Journal of Translational Medicine, researchers systematically summarize how the metabolic reprogramming of regulatory T cells within the tumor microenvironment maintains their immunosuppressive phenotype in colorectal cancer. The authors note that colorectal cancer represents a major global health burden, with its high mortality closely associated with tumor immune evasion driven by these regulatory cells.

According to the review, regulatory T cells adapt to the hypoxic and nutrient-deprived tumor microenvironment through coordinated metabolic pathways, including glycolysis, fatty acid oxidation, amino acid metabolism, the pentose phosphate pathway, and the PI3K-AKT-mTOR signaling cascade. Furthermore, the gut microbiome and its metabolites, such as short-chain fatty acids and bile acids, exert distinct regulatory effects on regulatory T cell metabolism within colorectal tumors.

Targeting these specific metabolic dependencies presents innovative therapeutic strategies to attenuate regulatory T cell-mediated immunosuppression. The authors evaluate the synergistic potential of combining metabolic interventions with existing immunotherapies, including immune checkpoint inhibitors and adoptive cell therapies, while proposing optimized clinical trial designs to address existing translational challenges and guide future research directions.
