In a randomized trial published in Genes & Nutrition, supplementation with a synergistic polyphenol blend modulated immune response pathways in runners following a half marathon. To evaluate specific innate immune pathways that may be improved by supplementation, researchers randomized participants to receive either a synergistic polyphenol blend (SPC; 1000 mg/d of standardized curcuminoids and pomegranate ellagitannins; Restoridyn®; Verdure Sciences; n = 13) or a placebo (n = 14) for 28 days prior to running a half marathon. Venous blood samples were collected before the run (PRE) as well as two hours (2H) and four hours (4H) post-exercise, with isolated RNA analyzed for the expression of 774 mRNA associated with 40 immune response pathways using a standardized panel.
At two hours post-run, participants receiving SPC exhibited attenuated activation of T-cell Activation and NK Cell Cytotoxicity, blunted activation of Type 1 IFN, and reduced suppression across JAK-STAT, Inflammatory Signaling, Lipid Metabolism, and Stress Response/DNA Damage pathways. Overall, the average differential expression change with SPC at two hours reflected 25% less suppression of immune pathways. By four hours post-run, the average differential expression change with SPC reflected a 232% enhancement of activation across multiple pathways, including IFN Signaling, JAK-STAT, Growth Signaling, Apoptosis Regulation, Metabolic Recovery, mTOR/Nutrient Sensing, and Transcriptional Regulation.
The study authors concluded that the observed SPC effect represents an improved immune responsiveness following a long-duration event, which may translate to faster resolution of inflammation and a return to normal function. While synergistic polyphenol blends containing curcumin and pomegranate ellagitannins had previously been reported to improve exercise recovery via reduced muscle injury and optimized inflammatory response, the authors noted that these molecular findings may have broad implications for individuals exposed to other types of physiological stress.
