Review maps key signaling pathways driving oral squamous cell carcinoma and targeted therapies

A new review published in Discover Oncology on 29 October 2026 examines the major molecular signaling pathways involved in oral squamous cell carcinoma (OSCC) and the targeted therapeutic strategies being developed against them. The article, authored by An Lou, Xuan Zhu, Zheming Li, Zhu Li, Xinjun Luo, Ming Li and Hongtai Pei, is an open-access review, meaning it synthesizes existing research rather than reporting new experimental results.

OSCC is described in the review as a prevalent head and neck malignancy, accounting for about 90% of oral cancers. According to figures the authors cite from the Global Cancer Observatory (GCO), the incidence of OSCC is projected to increase by approximately 40% by 2040. The disease can cause disfigurement and functional impairments, including difficulties with swallowing, speech and taste, significantly affecting patients’ quality of life and, in some cases, causing death. The authors argue that clarifying the molecular mechanisms behind the disease and developing more effective treatments are therefore important priorities.

The review’s central contribution is a structured survey of the key signaling pathways implicated in OSCC. These include the EGFR pathway, the PI3K/AKT pathway, the Wnt pathway, the JAK/STAT pathway, the MET pathway, the MAPK pathway, the PD-1/PD-L1 pathway, the Hippo pathway and the EMT pathway, among others. For each pathway, the authors say they examine its composition, the molecules involved in its regulation, and the inhibitory signals that can inactivate OSCC cells. They also connect these molecular mechanisms to clinical studies and to targeted therapies associated with each signaling route.

Among the targeted therapies highlighted for their therapeutic potential in treating OSCC are cetuximab, gefitinib and nivolumab. The authors note that these pathways are interconnected, forming networks rather than isolated linear chains.

The review concludes that understanding these interconnected signaling networks can accelerate the development of more effective and personalized therapies for OSCC. As a review article, it does not present new clinical trial data; its value lies in consolidating existing findings on pathway composition, regulation and drug targeting into a single reference for researchers working on the disease.
