PLA2G2A fuels gallbladder cancer stemness and immune suppression

A new study published in Cellular Oncology points to a single secreted enzyme, phospholipase A2 group IIA (PLA2G2A), as a multifunctional player in that process, acting on both the tumor cells themselves and the immune landscape surrounding them.

The work grew out of the authors’ earlier single-cell analyses, which had flagged PLA2G2A as a marker of early-stage gallbladder cancer and hinted at a possible role in regulating cancer stemness, the set of tumor-like stem cell properties that help cancers self-renew and resist treatment. The team, led by Mingye Gu, Zhengyuan Meng and colleagues with senior authors Hongyang Wang and Jing Fu, set out to test that hint directly.

Using gain- and loss-of-function experiments both in cell culture and in animal models, the researchers measured three cancer-relevant traits: stemness, tumor-initiating capacity and resistance to the chemotherapy drug gemcitabine. Raising PLA2G2A increased expression of the stemness-associated markers CD44 and CD133, boosted the cells’ ability to seed tumors and made them more resistant to gemcitabine.

Clinical sample data reinforced the laboratory findings: PLA2G2A expression correlated positively with CD44 and CD133 levels in gallbladder cancer specimens, linking the experimental mechanism to patterns seen in patient tumors.

The team then traced the signaling path downstream of the enzyme. PLA2G2A activates its receptor, PLA2R1, and promotes that receptor’s interaction with filamin A (FLNA), a cytoskeletal scaffolding protein. This receptor-scaffold partnership triggers ERK1/2 signaling, a well-known growth pathway, which in turn switches on stemness-associated gene expression. The authors mapped these connections using co-immunoprecipitation, western blotting and functional inhibition assays that blocked individual steps in the chain.

Beyond its effects inside tumor cells, PLA2G2A appears to reshape the tumor microenvironment. The researchers assessed the consequences for immune cells directly, finding that the secreted molecule helps sustain an immunosuppressive environment around the tumor.

Patterns in clinical samples matched this immune picture. High PLA2G2A expression correlated with infiltration by exhausted CD8-positive T cells, M2-polarized macrophages and regulatory T cells, the populations most associated with blunted anti-tumor immunity.

Taken together, the results position PLA2G2A as a driver that works on two fronts at once: it equips gallbladder cancer cells with stem-like, treatment-resistant behavior through the PLA2R1-FLNA-ERK1/2 axis, and it remodels the surrounding immune setting through soluble PD-L1. The authors describe this dual role as clarifying the enzyme’s place in gallbladder cancer progression.

The study, released as an open-access article on 1 October 2026, is being shared early as a peer-reviewed, accepted manuscript carrying a permanent DOI, with a final version of record to follow. The authors frame the findings as a rationale for further investigating PLA2G2A’s therapeutic potential, while noting that the work establishes mechanism and association rather than a tested treatment. Any move toward targeting the enzyme or its downstream signaling in patients would require the clinical studies that this preclinical and correlative evidence is intended to motivate.
