Four-gene model predicts biochemical recurrence in prostate adenocarcinoma in integrated study

Machine learning and neddylation biology reveal four prognostic genes in prostate adenocarcinoma

An integrated study combining public-dataset analyses with cell-line and mouse experiments identified four neddylation-related genes—TOP2A, SNRNP70, NFE2L2 and CAV1—and developed a prognostic risk model for biochemical recurrence in prostate adenocarcinoma. The report, summarized in a BMC Cancer abstract, combined differential-expression analysis, Cox regression and machine-learning algorithms to construct the model from patient datasets.

In the study’s reported evaluation, the four-gene model predicted biochemical recurrence in prostate adenocarcinoma. The abstract does not provide sample sizes, model-performance metrics, details of an independent validation cohort or evidence of prospective clinical utility. The findings therefore describe a computational prognostic association rather than a validated clinical test. Immune-infiltration analysis also found significant differences in five immune-cell types, including M1 macrophages, while single-cell RNA sequencing identified basal epithelial cells as a key population. SNRNP70, NFE2L2 and CAV1 showed distinct expression patterns between prostate adenocarcinoma and control samples.

The study’s Mendelian-randomization analysis reported associations between two candidate genes and benign prostatic neoplasm. CAV1 was associated with lower odds of the outcome, with an odds ratio of 0.275 and P = 0.037, whereas NFE2L2 was associated with higher odds, with an odds ratio of 5.154 and P = 0.007. These results are associations reported by the analysis; the abstract does not establish that either gene directly causes recurrence, changes tumor behavior or would guide clinical decisions.

Experimental work used reverse transcription-quantitative PCR in prostate adenocarcinoma cell lines and TGMAP transgenic mouse model tissues. TOP2A and SNRNP70 were significantly upregulated, while NFE2L2 and CAV1 were downregulated in both the cell lines and allograft tissues. In cultured prostate adenocarcinoma cells, pharmacological inhibition of neddylation with MLN4924 repressed TOP2A and SNRNP70 transcription and induced CAV1 expression. NFE2L2 messenger RNA levels remained stable, although its encoded NRF2 protein accumulated at the post-translational level.

Overall, the abstract describes a mixed study design: computational analysis of public patient data, exploratory immune and single-cell analyses, Mendelian-randomization estimates, and preclinical validation in cells and a transgenic mouse model. The findings nominate a study-defined prognostic signature and experimental candidates for further investigation, but the available abstract does not establish clinical readiness, clinical benefit or performance in prospective patients.
