Shared Genetic Architecture Links Fat Distribution, Ectopic Lipid Buildup and Metabolic Aging

A multivariate genomic analysis published in Biogerontology has constructed a latent genetic phenotype of adipose tissue aging and found that fat distribution across the body, ectopic lipid deposition in the liver and adipose-related metabolic signals share a common genetic dimension. The researchers integrated genome-wide association summary statistics for seven adipose- and metabolism-related traits and derived a composite genetic factor they named mvAdiposeAge, then mapped its signals across genes, pathways and tissues.

The team, led by Mingkai Li, Yunshu Yao and colleagues with senior author Haifeng Zhao, drew on summary statistics for visceral adipose tissue volume, abdominal subcutaneous adipose tissue volume, thigh subcutaneous adipose tissue volume, liver proton density fat fraction, fasting insulin, adiponectin and leptin. Rather than treating each trait in isolation, they applied Genomic Structural Equation Modelling, a statistical framework that estimates how genetic effects on individual traits load onto shared latent factors.

This modelling supported a shared genetic dimension linking adipose distribution, ectopic lipid accumulation and adipose-related metabolic traits. From that shared structure the researchers derived mvAdiposeAge, which acts as a genetically informed composite index: it captures the portion of genetic variation common across fat depot volumes, liver fat content and metabolic markers, rather than variation unique to any single measurement.

With the latent phenotype defined, the team ran a genome-wide association analysis against mvAdiposeAge and characterised its signals through FUMA-based functional annotation, MAGMA gene-level analysis, transcriptome-wide association analysis, expression fine-mapping, and pathway and tissue enrichment analyses. They also applied Mendelian randomization to probe relationships between the latent phenotype and related traits.

The genome-wide analysis produced a prominent signal at chromosome 16q12.2, with the variant rs1421085 in the FTO region emerging as the lead signal. MAGMA gene-level testing identified 49 genes that reached significance after false discovery rate correction.

Transcriptome-wide association analysis, which links genetic association signals to predicted gene expression, prioritised 11 genes. Expression fine-mapping with the FOCUS method further highlighted expression-mediated signals involving FTO, RPGRIP1L and AKTIP, although the study’s interpretation of these genes rests on the enrichment and fine-mapping results rather than new experimental validation.

Functional enrichment of the mvAdiposeAge signals implicated neurotrophin signalling, synaptic organisation, cAMP signalling, immune and phagocytic processes, endocrine regulation and metabolic pathways. Tissue enrichment analysis added a spatial dimension, suggesting that the signals were most prominent in specific brain regions. The analysis itself is genetic and does not demonstrate a mechanism connecting these pathways to adipose tissue aging.

The Mendelian randomization analyses supported positive associations of adiposity-related traits with mvAdiposeAge, and an inverse association between the latent phenotype and cognitive performance. These results describe directional genetic relationships estimated from instrumental variables, and they remain estimates that depend on the validity of the genetic instruments rather than proof of causation in the way a controlled experiment would provide.

Taken together, the authors present mvAdiposeAge as a latent genetic factor capturing shared variation across adipose-related phenotypes relevant to aging, and as a framework for further investigating the systemic biological associations of adipose tissue aging. Because the study is built entirely on genome-wide association summary statistics, its findings are associations at the genetic level rather than measured aging trajectories in individuals, and follow-up work in cohorts and experimental systems will be needed to test the biological pathways the analysis prioritises.
