MiR-323b-5p regulates the development of childhood asthma by targeting the RNF24 gene

Tiny RNA Molecule Emerges as Driver and Diagnostic Clue in Childhood Asthma

A small observational study involving 76 children with asthma and 73 healthy controls found that the microRNA miR-323b-5p was elevated in pediatric patients, while its putative target gene, RNF24, was reduced. The abstract also reports that miR-323b-5p showed diagnostic potential and was independently associated with childhood asthma in the analyzed group. These findings do not establish clinical diagnostic performance or a causal role in human disease.

According to the study abstract, investigators measured miR-323b-5p, RNF24 and α-SMA expression in serum and human airway smooth muscle cells using RT-qPCR, and measured cytokine levels with ELISA. They used Pearson correlation and logistic regression to assess clinical associations. The cell experiments examined proliferation and migration with CCK-8 and Transwell assays. Human airway smooth muscle cells were treated with PDGF-BB to establish an activated model for these experiments.

In the cell model, miR-323b-5p was significantly elevated in PDGF-BB-induced human airway smooth muscle cells, whereas RNF24 was downregulated. Inhibiting miR-323b-5p reduced cell proliferation, migration and the measured inflammatory response in this experimental system. The reported suppressive effects were reversed when RNF24 was co-knocked down, which the authors interpret as evidence that miR-323b-5p acts through RNF24. These mechanistic results are based on cultured cells and do not demonstrate a clinical treatment effect.

The research, summarized here from the available abstract, suggests that miR-323b-5p may warrant further evaluation as a biomarker and as a possible therapeutic target in childhood asthma. The study’s scope is limited: the human comparison was small, only the abstract was available for this report, and the cellular findings cannot by themselves establish how miR-323b-5p and RNF24 contribute to asthma in patients. No outcome from a clinical miR-323b-5p intervention was reported.
