Review Proposes Age Decompression Framework for Cancer Risk in Lung Cohorts

Share on FacebookShare on TwitterShare on LinkedinShare on RedditShare on Telegram

Secondary analyses using public lung adenocarcinoma cohorts demonstrate that chronologic age explains little of the interindividual variation in tumor and microenvironmental states, despite retaining prognostic information after adjustment for routine clinical factors. In a review published in Discover Oncology, researchers synthesize evidence from aging biology and geriatric oncology to evaluate how chronologic age functions in cancer risk stratification and clinical decision-making, reporting that the risk sources carried by age are multidimensionally heterogeneous.

The authors state that chronologic age remains an important reference in oncology but functions fundamentally as a composite variable aggregating multiple biological processes and clinical factors. Consequently, chronologic age retains predictive value at the population level while providing limited biological explanation at the individual level. To address this limitation, the authors propose an age-decompression framework to examine age-associated risk across host aging, tumor evolution, ecological selection, and related clinical factors.

This framework is not intended to replace chronologic age in clinical practice. Instead, it seeks to identify the underlying basis of its predictive value by repositioning chronologic age from a single biological proxy to a clinical risk signal that requires further resolution, aiming to move research on cancer and aging from age-based prediction toward mechanistic understanding.

Subject of Research: Mechanisms of chronologic age as a clinical risk signal in oncology

Article Title: Decompressing entangled mechanisms within chronologic age defines the true value of age as a clinical signal in oncology
