Study maps phosphorylation network linking MLLT1 to transcriptional control in cancers

Researchers have carried out an integrative bioinformatic analysis of MLLT1, a chromatin-associated protein better known as ENL, to map how its phosphorylation may connect to transcriptional regulation in cancers.

MLLT1 sits within the super elongation complex (SEC), a molecular machine that drives transcriptional elongation. The protein is frequently implicated in leukemias and in Wilms tumor through fusions and mutations. According to the authors, the phosphoregulatory mechanisms of MLLT1 had remained underexplored, which motivated the new analysis. The team mined large-scale phosphoproteomic datasets to identify predominant phosphorylation sites on MLLT1, pinpointing three: S292, S359 and S361.

From these phosphosites, the researchers constructed co-phosphoregulated protein networks using Fisher’s Exact Test, Gene Ontology enrichment analysis and protein interaction databases. The resulting network was enriched in chromatin remodeling, histone binding, transcription elongation via the SEC and PAF complexes, RNA processing, and genome stability. Among the three sites examined, S292 emerged as a significant phosphorylation site in the analysis.

The analysis also pointed to upstream kinases and interaction partners that may tie MLLT1 phosphorylation to epigenetic control. CDK9 was identified as an upstream kinase, while binary interactors included KMT2A and DOT1L, both well-known players in chromatin and transcriptional regulation. The authors report that these findings underscore how MLLT1 integrates phosphorylation with epigenetic mechanisms.

When the phosphosites were linked to cancer associations, the analysis connected them to leukemia and Wilms tumor, and suggested potential links to lung cancers as well. The authors state that these associations highlight novel therapeutic avenues, although the work is a computational, dataset-based analysis rather than an experimental or clinical study, and the reported cancer links are associative in nature.

Overall, the study provides mechanistic insights into how MLLT1’s phosphoregulated functions may contribute to transcriptional control and oncogenesis, according to the abstract. Because the article is being shared early as a peer-reviewed, accepted manuscript, it remains subject to further edits before the final version of record is published.
