New Graph Pangenome Pipeline Simplifies Plant Organelle Genome Analysis

Share on FacebookShare on TwitterShare on LinkedinShare on RedditShare on Telegram

Researchers have introduced MineGraph, an evolutionary graph pangenome (evograph) pipeline designed to streamline diversity analyses of plastid and mitochondrial genomes across species and families. The tool addresses a growing challenge in genomics: as sequencing output expands, graph pangenomes are increasingly used in evolutionary studies, but computational costs and complex interpretations at higher taxonomic levels have limited their broad adoption.

To demonstrate the framework, the team applied the pipeline to 709 chloroplast genomes from the plant order Poales. The analysis indicated that genomic diversity in this group is driven primarily by species-specific variation, a finding the authors report in the abstract of their study.

The pipeline resolved a 77.7 Kbp consensus genome for the Poales dataset and applied graph theory metrics to the data. According to the study, these metrics highlight distinct genus-level evolutionary signatures, suggesting that evographs can capture evolutionary complexity across high taxonomic levels in a nuanced way.

The article, published in Genome Biology on 21 October 2026 by Rakan Haib and Sariel Hübner, is open access. The authors position MineGraph as a practical route for researchers who want to use graph pangenome approaches in evolutionary studies without the computational burden and interpretive complexity that have hindered wider use at higher taxonomic levels. The reported results are based on the Poales chloroplast dataset described in the study abstract; the published version is an early-release, peer-reviewed accepted manuscript that remains subject to further edits before the final Version of Record.
