Proteomic Study Identifies Over 5,000 Proteins Across CALHM6 Comparison Groups in Vitro

A mass spectrometry-based proteomic investigation has identified 5,282 proteins across comparison groups in cellular models evaluating the regulation of Calcium Homeostasis Modulator Protein 6. The in vitro study analyzed differentially expressed proteins under both plasmid-mediated overexpression and small interfering RNA knockdown conditions to investigate the target protein’s functional roles.

To establish the experimental models, the researchers confirmed CALHM6 overexpression and knockdown at the messenger RNA level using real-time polymerase chain reaction. Protein-level alterations were subsequently verified through western blotting prior to downstream proteomic measurements.

Cell lysates were collected for proteome analysis twenty-four hours post-treatment before undergoing mass spectrometry analysis. The resulting proteomic datasets encompassed three comparison sets: overexpression versus siRNA control, knockdown versus siRNA control, and overexpression versus knockdown.

Computational evaluation of the identified differentially expressed proteins was conducted using Python data analysis workflows alongside pathway enrichment through the Metascape platform. These analytical tools were applied to examine the pathways and proteins identified across the overexpression and knockdown groups.

CALHM6 is a tetraspanin-like membrane glycoprotein belonging to the family with sequence similarity 26, member F, that contributes to regulating gene expression and cellular processes. While prior evidence supports a role as a calcium modulator with potential immunomodulatory functions in infections and cancer, the underlying signaling cascades remain largely unknown, with the current findings shedding light on molecular mechanisms governing its immunomodulatory activities.
