The scaffold protein AXIN1, long known as a core component of the Wnt signaling pathway, has now been shown to play two distinct roles during cytokinesis: first, as part of a β-catenin p-S60-dependent machinery that drives cleavage furrow ingression in early telophase, and second, as a midbody ring component that orchestrates the sequential recruitment of the ESCRT complex during abscission in late telophase. Depletion of AXIN1 in HeLa cells caused ECT2 delocalization, reduced RHOA activity, and a dramatic rise in multinucleated cells and cleavage furrow regression, revealing a previously unreported function of this tumor suppressor at the Flemming body.
